Can prenatal PFOS exposure alter brain reward circuits and increase addiction risk?

What's actually in it

PFOS (perfluorooctane sulfonate) is one of the most common PFAS chemicals. It was used in nonstick coatings, stain-resistant fabric treatments, and food packaging. Although production has been phased out, PFOS persists in the environment and in people's blood for years. It crosses the placenta readily, exposing the developing fetal brain.

What the research says

A 2026 study in Neurotoxicol Teratol investigated how developmental PFOS exposure affects gene expression in the nucleus accumbens, the brain region that processes reward, pleasure, and motivation.

Prenatal PFOS exposure altered the expression of genes involved in dopamine signaling, synaptic plasticity, and glutamate transmission. These are the same pathways hijacked by addictive substances. The changes made the reward circuit more sensitive to stimulation, a pattern associated with higher addiction vulnerability.

The study also found changes in genes that regulate stress response and impulse control, both of which affect susceptibility to substance use disorders.

These effects were observed at PFOS levels within the range found in the general population, not just in heavily contaminated areas. The brain changes were established before birth and persisted into adulthood.

While these findings don't mean PFOS directly causes addiction, they suggest prenatal exposure may set the stage for higher risk. Reducing PFAS exposure before and during pregnancy through safer cookware, filtered water, and uncoated food packaging helps protect fetal brain development.
